Abstract

Trentepohlia sp. (TP1) was collected from tree barks at Kodaikanal, Tamil Nadu, India.
Chloroform and methanol (7:3) obtained the maximum amount of carotenoid pigment (5.72 %)
and there is a difference in the Rf values of carotenoid pigment extracted with solvents (0.95), and
coconut oil (0.67) by TLC. Petroleum ether and acetone (9:1) were used as a mobile phase in
column chromatography and elution 2 yielded 64.096±0.17 mg/mL of carotenoid. The GC-MS
and FT-IR analysis revealed that about 90% of canthaxanthin was obtained from the purified
carotenoid pigment. The concentration of canthaxanthin was inversely proportional to the free
radicals generated in the DPPH free radical scavenging assay. The IC50 values of canthaxanthin
from Trentepohlia sp. (TP1) was 23.37±1.52 µg/mL, which was close to the positive control
Ascorbic acid (22.49±0.43 µg/mL). This study reveals the prospective approach on production of
canthaxanthin from Trentepohlia sp. using a novel solid-solid-state twin-layer photobioreacto
